Finishing Drosophila mojavensis Fosmid Clone 485-B16

چکیده

With the sequences of so many organisms’ genomes finished and available for use, comparative genomics has become a rapidly growing scientific discipline. In the spring of 2007, we plan to use comparative genomics to study the 4 chromosome (the ‘dot’ chromosome) of Drosophila by comparing the sequence of the Drosophila mojavensis dot chromosome to the sequences of the Drosophila melanogaster and Drosophila virilis dot chromosomes. Although this chromosome appears to be mainly heterochromatic in D. melanogaster and euchromatic in D. virilis, Slawson et. al showed that it contains genes. We hope to learn about the D. mojavensis dot chromosome from sequence comparisons, but first the sequence of the D. mojavensis dot chromosome must be finished. This paper describes the first step in the comparative genomics work to be accomplished in the spring of 2007, the finishing of a D. mojavensis dot chromosome fosmid to mouse standard.

Finishing Drosophila mojavensis Fosmid Clone DMAC-1a

Bio 4342 students are currently working on finishing the Drosophila grimshawi dot chromosome and a euchromatic region of Drosophila mojavensis. Project DMAC-1a (from D. mojavensis) began in two contigs with only a single macroscale problem and a few microscale problems. Using Consed and phredPhrap, this project has been finished to completion and is now in a single contig. This paper presents t...

Finishing Drosophila virilis Fosmid Clone 4N16

The overarching goal of Bio 4342/W research over the past several years has been to understand a euchromatic region of the Drosophila virilis genome well enough to be able to distinguish this domain at the DNA level from the heterochromatic counterparts in its genetic relatives, such as Drosophila melanogaster, the common fruit fly. The class will utilize the well-established genome of D. melan...

euchromatic. Obtaining high quality sequences for both species will enable comparative analysis of chromosome four, providing sequence information about the basis of gene packaging. My project contributes to the final sequence data of D. virilis by finishing fosmid 14p24

Chromosome four of the Drosophila species melanogaster and virilis is of interest to investigators because the first is mostly heterochromatic while the second is mostly euchromatic. Obtaining high quality sequences for both species will enable comparative analysis of chromosome four, providing sequence information about the basis of gene packaging. My project contributes to the final sequence ...

The small fourth chromosome of Drosophila melanogaster, also known as the dot chromosome, is unlike the other three chromosomes of Drosophila in that it contains largely heterochromatic sequence. The 1.2 Mb arm of the dot chromosome has normal gene density, but a higher frequency of repetitive sequences compared to the euchromatic arms of chromosomes 13. The many genes packaged in the dot chrom...

Molecular genetic characterization of a locus that contains duplicate Adh genes in Drosophila mojavensis and related species.

Drosophila mojavensis and other species of the mulleri subgroup contain a duplicate gene encoding the enzyme alcohol dehydrogenase (ADH). Studies on the genetic relationship of the two genes using electrophoretic variants show them to be closely linked. We have cloned a 13.5-kb fragment of D. mojavensis DNA into the lambda vector, Charon 30. This fragment contains both Adh genes separated by ap...
